Juan Musso began his football career at Club de Regatas San Nicolas’ youth academy. He was quickly loaned out to Defensores de Belgrano de Villa Ramallo, where he won the local title.

After that, he enrolled in the Racing Club de Avellaneda, or simply Racing Club. He was promoted to the first team in 2016 after spending years at the academy and playing for younger teams. However, because he wasn’t the team’s first-choice goalkeeper, he didn’t make his debut until May 28, 2017, in a 2–1 home victory over San Lorenzo.

He began to improve his performance and put in long hours of training to eventually become the club’s starting goalie. In 2018, he was sold to Serie A club Udinese Calcio, aka Udinese, after two seasons with the main squad and 36 appearances.

His reported transfer fee was €4 million. During his three seasons at the club, he managed to establish himself as a crucial player. He appeared in 104 games for the team throughout those three seasons.

On July 2, 2021, he left Udinese after signing with another Serie A team, Atalanta Bergamasca Calcio, or Atalanta. It was reported that his transfer cost €20 million. Since then, he has made 45 appearances for Atalanta.

In 2013, the San Nicolás native was called to his first national team camp as he was named to Argentina’s under-20 squad for the preparation of the 2013 South American Youth Football Championship.
